A Novel Host Defense System of Airways Is Defective in Cystic Fibrosis: UpdateTo the Editor:We have recently discovered that the 5-thio-2-nitrobenzoic acid (TNB)based hypothiocyanite (OSCN) detection assay is affected by the presence of H2O2. As we had not removed H2O2 prior to the TNB assays (the results of which were reported in Figures 3 and 5 of our recent AJRCCM article [1]), we reexamined the rates of OSCN production of primary airway epithelia and tracheal explants. Following catalase pretreatment to scavenge H2O2, the TNB method detected rates of OSCN production very similar to those reported in Figures 3 and 5, indicating that H2O2 was indeed effectively utilized for hypothiocyanite generation in our system. This methodological correction does not influence the conclusions of our article, but is an important consideration for scientists actively engaged in research in this field.
